What is Rowan GLP3(R)?

GLP-3(R) is a research analogue of retatrutide — a triple agonist that targets GLP-1, GIP, and glucagon receptors simultaneously. Adding glucagon receptor agonism to the dual incretin approach introduces a third mechanism: glucagon drives hepatic fat oxidation, thermogenesis, and energy expenditure. A Phase II clinical trial published in the New England Journal of Medicine in 2023 showed a mean body weight reduction of 24.2% at 48 weeks at the highest dose — the largest reduction reported for any single compound in a clinical trial at the time.

The 2023 NEJM Phase II trial of retatrutide showed 24.2% mean body weight reduction at 48 weeks — the largest single-agent weight reduction ever reported in a clinical trial at the time. GLP-3(R) is the research analogue for studying how adding glucagon receptor agonism produces this step-change result.

How GLP-3(R) works in research models

GLP-3(R) is a triagonist targeting GLP-1, GIP, and glucagon receptors simultaneously. The addition of glucagon receptor agonism to the incretin dual agonist framework adds hepatic fat oxidation (β-oxidation), thermogenesis, and increased energy expenditure — mechanisms typically suppressed by GLP-1 agonism alone.

The addition of glucagon receptor co-agonism to the GLP-1/GIP framework adds hepatic fat oxidation and thermogenesis — pathways typically suppressed by GLP-1 agonism alone. Together, the three receptors address appetite, metabolism, and energy expenditure simultaneously.

Questions & Answers

You ask, we answer.

Paradoxically, no. Although glucagon raises blood glucose, when combined with GLP-1 agonism (which drives compensatory insulin), the net glycaemic effect is neutral. The glucagon component instead drives liver fat oxidation, thermogenesis, and increased energy expenditure — additive to the appetite suppression from GLP-1 and GIP.

The Phase II NEJM trial of retatrutide (2023) showed 24.2% mean body weight reduction at 48 weeks at the highest dose — the largest reduction reported for any single therapeutic agent in a clinical trial at the time of publication.

GLP-3(R) is a research analogue with the same triple GLP-1/GIP/glucagon receptor agonist mechanism as retatrutide (Eli Lilly's LY3437943). It is not the approved pharmaceutical product and is supplied exclusively for laboratory research use only.

Dual agonism (GLP-1 + GIP, the tirzepatide mechanism) showed ~20-22% weight loss in clinical trials. Triple agonism adds glucagon receptor activation, which contributes hepatic fat oxidation and thermogenesis on top of the dual incretin effects. Phase II data suggests this additional mechanism produces a further 2-4% mean weight reduction.

The triple agonist mechanism is the frontier of this research area. Glad someone is carrying it with real documentation. Vial arrived sealed and intact, COA matches. This is the brand for serious research.

Running a comparison research protocol across the GLP series. Having all three from the same vendor with the same testing standard is something I didn't think I'd find. Rowan made it easy. GLP3(R) arrived in the same quality as the others.

Triple receptor agonist research is still early stage in the literature but the mechanism is compelling. Rowan is one of the only vendors even carrying GLP3(R) with verified purity. The COA showed 99.0% which is exactly what I needed to feel comfortable proceeding. Fast ship, clean vial.
